Setup: Intonation advise needed

3 things:
intonation
pickup height
adjustment of the trem

just got this am std strat and would like to get it nice and playable.
how much would this setup cost?
 
So to check intonation, the tuning for the 12th fret and 12th fret harmonic must be the same? Harmonic is simply placing the string on the fret metal right? And bring the saddle left is to bring the tuning lower?

No, my point is,

For intonation

How is NOT using harmonics

more accurate than

using an open string alone


against the fretted note.

To my understanding, the harmonic is more accurate, and I have never heard of reputable techs/luthiers who do NOT use the harmonic.
 
The fretted note is more important, because that is the note you play. If you fret harder, the note will be sharper unlike the harmonic which doesn't require you to fret. How well a tech can set intonation depends on how well he understands how hard one presses.
 
Thats correct (duh :P) GC, but perhaps you'd like to comment on this.

The reason the harmonic (12th fret, or whatever harmonic technique) is used, is to ensure the string is as in tune as possible; its used as a point of comparison with regards to the fretted 12th fret note with intonating.

So, how does NOT using the harmonic, but just tuning the string alone (as in Taylorboy87's comment, of which i assume he tunes his string first) and then intonating, fare against using the harmonic to tune? Which is more accurate?
