google 'cigar box guitars'. There's a whole counter-culture out there on it.

I built a cigar box guitar once, and it sounds great. You'll find instructions on the sites on how to do it - including making your own pickup

If you actually want to physically build a guitar, rather than assemble one (big difference btw), then start with a guitar box guitar. It'll probably be the easiest way to start.
